چکیده

Gallbladder carcinoma is an uncommon, but highly malignant tumor, with poor prognostic, and diagnostic manifestations in early stages. The Indian Council of Medical Research reported increased incidence of gallbladder carcinoma in the surviving population of the Bhopal gas tragedy that involved exposure of more than 500,000 people to methyl isocyanate gas. The severity of exposure, and increased multi-systemic morbidity in the survivors stimulated us to examine the molecular changes leading to gallbladder carcinoma. Surgically resected samples (N = 40) of gallbladder carcinoma were studied for the p53, Rad50, and cyclin-E expression by immunohistofluorescence bioimaging. Among the 40 samples, 23, 11, and 10 showed p53, Rad50, and cyclin-E expression, respectively, in moderately differentiated adenocarcinomas, demonstrating the prevalence and invasiveness of this disease in the methyl isocyanate-exposed population (P = 0.0009). Nevertheless, co-expression of Rad50, and cyclin-E with p53 was absent in adenomas with dysplasia, demonstrating their independent roles. We conclude that there was altered expression of p53, Rad50, and cyclin-E in the malignant transformation of gallbladder carcinoma in this methyl isocyanate gas-exposed cohort. Hence, these proteins may be useful as markers to identify premalignant lesions that are likely to progress into malignant adenocarcinoma.

Frequency of genetic alterations observed in cell cycle regulatory proteins and microsatellite instability in gallbladder adenocarcinoma: a translational perspective.

Genetic alterations are considered to play an important role in both biological behavior and progression of human malignancies. However, cancer of gallbladder (CAGB) is an obscure phenomenon and is highly malignant with underprivileged diagnosis and poor survival (Chan et al., 2003). Earlier, we reported a significant higher prevalence of CAGB amongst victims of Bhopal gas tragedy exposed to me...

تغییرات بیان ژنهای P53 ، Cyclin-D1 ، RB1 ، c-Fos ، N-ras در هپاتوسلولار کارسینوما در ایران

Background: Hepatocellular carcinoma is the most common primary malignant tumor of the liver. The effect of some genes especially those involved in cell cycle regulation have been shown in the development of this cancer in several studies but there are some controversies about them yet. Materials and methods: The paraffin-embedded tissue samples of 25 patients (18 males and 7 females) with hep...

An immunohistochemical study of the expression of cell-cycle-regulated proteins p53, cyclin D1, RB, p27, Ki67 and MSH2 in gallbladder carcinoma and its precursor lesions.

Gallbladder carcinomas are rare but highly lethal neoplasms. We examined the expression of five cell-cycle-related molecules (p53, RB, cyclin D1, p27, Ki-67), and MSH2, in 46 carcinomas, 14 adenomas, 15 low-grade dysplasias, 9 intestinal metaplasias and 20 normal gallbladder epithelia. The expression of these molecules was altered in gallbladder carcinomas and adenomas. In gallbladder carcinoma...

A preliminary study of microsatellite instability analysis in different genotypes of p53 codon 72 in breast invasive ductal carcinomas

Abstract   Background: The polymorphic variants at codon 72 of the p53 gene, encoding either   proline or arginine at residue 72, produce marked change in the structure of p53.   From the evidence that the DNAmismatch repair system and p53 interact to maintain   genomic integrity, we hypothesized that the codon 72 variation may influence the   prevalence of microsatellite instability a featur...

سنجش فراوانی بیان پروتیین‌های p53 ، Ki67، CD99 و Fli-1 در بلوک‌های پاتولوژی یووینگ سارکوما

Background: Ewing sarcoma family tumors (ESFTs) are among the most malignant tumors in children and young adults. ESFTs include Ewing sarcoma (ES) and peripheral primitive neuroectodermal tumors (pPNETs). As there seemed to be few studies on the molecular biology of ESFTs, we investigated the frequency of CD99, Ki67, p53 and Fli- 1 protein expression in 15 Iranian patients with ESFTs. In addi...
